abstract

abstract background & objective to assess the extent of l. monocytogenes in causation of human spontaneous abortions by isolation methods and pcr analysis for the presence of acta and inlb genes. methods : in this descriptive study, vaginal swabs were collected from 96 women with spontaneous abortions who referred to tehran`s hospitals. the samples were cultured in to listeria specific media (palcam agar). then , the listeria genus was verified by differentiation  biochemical tests, such as, haemolysis on blood agar, catalase and oxidase reaction, motility at room temperature. pcr test was performed on all samples and detected the acta and inlb genes of l. monocytogenes. results : in culture, 7 of 96 samples were positive for l.monocytogenes.                                     in pcr technique, acta and inlb genes were detected from 12 and 2 vaginal samples,  respectively. conclusions : the occurrence of pathogenic l. monocytogenes in cases of spontaneous abortions was 12.5%. it seems that the acta and inlb genes have an important roles as essential virulences determinant in pathogenic l.monocytogenes. the results show the pcr method is more sensitive, easier and faster than culture to detect l.monocytogenes. key word : listeria monocytogenes , spontaneous abortions , acta gene , inlb gene

Determination of Dominant Serovars and Molecular Analysis of hly and iap Genes Related to Listeria monocytogenes Strains Isolated from Spontaneous Human Abortions in Tehran

Background and Aims: Listeria monocytogenes, a gram positive, facultative, intracellular bacterium is the causative agent of listeriosis that is transmitted to human through raw and ready-to-eat foods. The aim of the present study was to determine dominant serovars of L. monocytogenes isolated from spontaneous abortion using phenotypic and genotypic methods. Materials and Methods: In present s...

Investigating the prevalence of resistance genes in Listeria monocytogenes isolated from food and clinical samples

Background: Listeriosis can be fatal for vulnerable groups of society. The disease has been widespread in recent years due to the large consumption of dairy and meat products. There is little information about the susceptibility of antibiotics and the pattern of Listeria monostigenesis gene resistance in Iranian society. Accordingly, the present study was conducted to investigate the antibiotic...

Distribution and Characterization of Dominant Serovars of Listeria monocytogenes Strains Isolated from Spontaneous Human Abortion in Tehran

Background and Aims: The aim was to determine the role of dominant serovars of Listeria monocytogenes (L. monocytogenes) in spontaneous abortions, using isolation methods and polymerase change reaction (PCR). Materials and Methods: A total of 258 samples comprising of placental tissue, vaginal swabs and blood were collected from 123 patients with spontaneous abortion. L. monocytogenes was iden...

Intracellular induction of Listeria monocytogenes actA expression.

Following entry into the host cytosol, the bacterial pathogen Listeria monocytogenes dramatically increases the expression of several key virulence factors. The expression of actA, whose protein product is required for L. monocytogenes actin-based intracellular motility, is increased by more than 200-fold in cytosolic bacteria in comparison to broth-grown cultures. Two distinct promoter element...

ActA Promotes Listeria monocytogenes Aggregation, Intestinal Colonization and Carriage

Listeria monocytogenes (Lm) is a ubiquitous bacterium able to survive and thrive within the environment and readily colonizes a wide range of substrates, often as a biofilm. It is also a facultative intracellular pathogen, which actively invades diverse hosts and induces listeriosis. So far, these two complementary facets of Lm biology have been studied independently. Here we demonstrate that t...

Mechanism of polarization of Listeria monocytogenes surface protein ActA

The polar distribution of the ActA protein on the surface of the Gram-positive intracellular bacterial pathogen, Listeria monocytogenes, is required for bacterial actin-based motility and successful infection. ActA spans both the bacterial membrane and the peptidoglycan cell wall. We have directly examined the de novo ActA polarization process in vitro by using an ActA-RFP (red fluorescent prot...
